Strangeways Nick went into lockdown for a week, with inmates locked in their cells unable to exercise or even shower.
HM Prison Manchester has banned convicts from leaving their rooms for any reason amid an “intelligence-led” maximum security operation.
Visits to the Category A prison, also known as the Stangeways, have been suspended since Monday.
Relatives told Manchester evening news how the inmates have no fresh air and food is brought to their door.
The Department of Justice (MoJ) said the lockdown stemmed from an “intelligence-led operation” that took place at the prison to crack down on illegal goods.
Officials confirmed visits have been temporarily suspended and are expected to resume this weekend.
But bosses have not confirmed when the lockdown will end.
A woman whose 19-year-old son is an inmate at the prison said the atmosphere inside had become “very tense.”
She said: “My son called me to say that the prisoner’s mental health was deteriorating rather quickly.
“They say they haven’t showered and get one meal a day delivered to their cell.
“He said they haven’t had any fresh air since last Wednesday.
“I know they’re all there for crimes, but they have human rights and it’s really not good for their mental health.”
The mum added: “It was bad enough but because of the holidays it will now be around eight or nine days with no visitors.
“He says prison officers went in to search people’s cells.”
Another woman, whose partner is also an inmate, said she was told prisoners had been locked up for four days without being able to shower or exercise.
She described how prisoners kicked their doors and alarms rang in the background when she called the prison.
Another man, whose friend is in jail, said: “They got a letter last week saying the jail is going into lockdown and it was expected it could be a week longer.
“He’s been behind his door for five days.
“The person I know said today will be her sixth day without a shower.
“Prison guards bring meals to their cell and won’t even let them out for dinner. He said his head was starting to walk.
“Certainly they have basic human rights and should be able to shower.
“They weren’t even let out to exercise. They have a cell phone system, so he could call me, but he can’t leave his cell phone.”
The MoJ commented on the week-long lockdown.
A spokesman said: “An intelligence operation took place this week to crack down on illegal smuggling that is endangering detainees and staff.
“Visits have been temporarily suspended but are expected to resume this weekend while phone calls to family and friends have been available throughout.”
